Our primary objective in this project is to leverage the knowledge acquired from our recent research on novel inhibitors of flavivirus and poxvirus replication. We aim to build upon the outcomes of both our fundamental and previous applied research to develop optimized compounds that meet the criteria for preclinical candidates, and with which we plan to conduct proof-of-concept experiments in murine models of diseases such as TBE and mpox. Our objective is to identify compounds capable of inhibiting the key replication enzymes of diverse flaviviruses, as well as modulating the immune response to prevent viral dissemination within the host. Our preliminary results indicate that certain methyltransferase inhibitors and STING agonists may also exhibit activity against a broader spectrum of viruses spanning several families.
Furthermore, we intend to explore potential combinations of our designed and synthesized derivatives with compounds that have demonstrated promising antiviral activity against complementary biological targets in our previous tests, thereby achieving a synergistic effect through agents with distinct mechanisms of action (e.g., a combination of RdRp and MTase inhibitors).
